Per Sinemet 25/100 tab Carbidopa 25 mg, levodopa 100 mg. Per Sinemet 25/250 tab Carbidopa 25 mg, levodopa 250 mg
Indications/Uses
Dosage/Direction for Use
Sinemet 25/100 Initially 1 tab tid. Increase by 1 tab everyday or every other day. Max: 8 tab. Sinemet 25/250 Initially ½ tab once daily to bid. Add ½ tab everyday or every other day. Max: 8 tab.
Administration
Should be taken on an empty stomach: May be taken w/ meals if GI discomfort occurs. 
Contraindications
Narrow-angle glaucoma. Undiagnosed skin lesions or history of melanoma. Hypersensitivity to any component of this product.
Special Precautions
Not recommended for drug-induced extrapyramidal reactions. CV diseases including history of MI or arrhythmias or pulmonary disease, bronchial asthma, renal, hepatic or endocrine disease, history of psychoses or convulsions or wide-angle glaucoma. History of active peptic ulcer. Absorption of levodopa may be impaired in some patients on a high-protein diet. Discontinue use at least 6-8 hr prior to anaesth. Childn <18 yr. Pregnancy, lactation.
Adverse Reactions
Dyskinesia including choreiform, dystonic & other involuntary movements. Mental changes including paranoid ideation & psychotic episodes; depression w/ or w/o development of suicidal tendencies; dementia; & GI disturbances.
Drug Interactions
Discontinue MAOIs 2 wk prior to therapy (except low doses of selective MAO-B inhibitors); TCA, antihypertensives, phenothiazines, butyrophenones, phenytoin, papaverine. Co-administration w/ milk.
